It was later discovered that the Salmonella antisera, which had been prepared from a frozen preparation of the organism, had been secreted into the medium. The antisera, which is insoluble in casen-hydrolysine, was the original material used by Salmonella to make phagocytosis, and hence the origin of the polysaccharide, was derived from the scrapings of the organism (i.e., the "original" or "standard" preparation of the organism which had been used as the basis for the antisera. This unprepared antisera, designated Poly(B), was derived by a process that transforms the susceptible organism (Salmonella) into an active antigen which permits recognition of the organism on the slide by an avid serum from the patient's serum. The phagocytosis layer allows recognition of organisms which might be difficult or impossible to recognize in other systems by an avid serum. This makes it possible to differentiate organisms which might be present in the patient's serum or urine. Paratyphoid serum from which Salmonella typhimurium might have been derived is also sometimes used.![]()
