Hanks' Balanced Salt (HBSS) is used for culturing gram-negative bacteria and non-gram-negative bacteria. Sodium chloride maintains osmotic balance in the solution, while magnesium and calcium maintain the acid-fast stability. HBSS is provided without, low molecular weight, deionized water and is low in bilirubin. Low molecular weight ions dominate in this medium, making it most suitable for use with the Klar family of bioluminescent indicator dyes, which reduces the solute pool. The varied ionic strength of these HBSS solutions, made without calcium and magnesium, allows selection of the most acid-fast and ion-reducing agents. Individually wrapped, sterile vials are sealed in polyethylene bags to prevent evaporation of the liquid during storage. The bags are gamma-irradiated to eliminate the risk of cell lysis.
