Not familiar with "Big Hole". Is this a standard M4 / AR15 upper receiver? Thank you

The Daniel Defense Big Hole Upper Reciever w Feed Ramps 04-012-09001 is designed and built with a .315 front pin hole to fit the early Colt lowers with .315 front holes.

Will this work with 300 blackout barrel?

yes it will. no difference from a milspec "small hole" upper.
It will only fit early model Colt SP1 lowers with a large hole front take down pin.
