will this co witness with a Sig Romeo pro 1?

These will be too short for the Romeo Pro 1. For that optic we recommend our 345-332 height.

Have to use a Holsun mount plate with the EPS Carry eliminates the use of the factory P365 iron sights. Will these sights work with the higher optic?

Yes, these sights will provide a lower 1/3rd co-witness on a Sig P365 w/ EPS Carry and Holosun mounting plate.

Will this co witness lower 1/3 with Romeo zero

This height will be a little taller than lower 1/3rd if you are using a Romeo Zero without a mounting plate. For a lower 1/3rd co-witness, we would recommend using our 240-238 height sights. You can find those by searching SIG-180-240-238 in the Optics Planet search bar.
