Are the 3 little air holes supposed to face upward?

Install it just as pictured. Compensation holes at the 3 and 9 o'clock positions. The small hole will be at the 1 o'clock position. Barrels naturally try to rise to the right due to the has force exerted on the ID of the bore. These holes allow an escape for that push force. This positioning is referred to as "clocking".

No. The POF USA Muzzle Brake And Collar Jam Nut will only be compatible with a 1/2x28 threaded barrel. You would need to have your barrel replaced with a threaded version or have your current barrel threaded for a muzzle device.
