Will the Holosun HS407K work with this mounting plate?

The Holosun 407K is not compatible with this optic plate. The 407K uses the RMSc/Deltapoint Pro mounting footprint. The correct plate for a Holosun 407K and optic ready VP9 can be found by using the link below:

EGW part number 49513 will fit a 507C and a Non-optics ready G22

No, this Evolution Gun Works adapter plate fits the optics ready VP9 and allows the user to fit an optic with an RMR footprint. If your slide already has an RMR cut you can fit any optic with the Trijicon RMR optic footprint.
