Yes, the LPA Adjustable Rear Shotgun Sights are compatible with the Benelli M4. As per the manufacturer: This rear Sight works on most Famous brands of Semi-Auto and Pump Shotguns such as Beretta, Benelli, Mossberg, Browning, Winchester, Ithaca etc... with barrel diameters ranging from .819" to .842" for standard shotguns, and .858" to .882" for heavier gauges. Center-to-center mounting hole distance: 1.7".

Will this fit the factory drilled holes on a Mossberg 590?

The LPA Adjustable Rear Shotgun Sights - Screw Down sights might work with a Mossberg 590. However, I recommend contacting the manufacturer to confirm compatibility.

No I dont believe so you will need a newer taller front sight either the SG2 or SG3 for that model Thanks again Fusion Firearms

It is suggested you use some sort of a front sight for a better sight alignment.
