No, this is not compatible with the Enhanced uppers that have the built in barrel nut.

Yes. It does. I just completed a build. You will not need the barrel nut that comes with the Spike's product though. It is removable. The Spike's provided barrel nut is only needed for standard (non-enhanced) uppers. After taking out the provided barrel nut, the quad rail attaches perfectly to the enhanced upper with the 8 screws that are provided. It looks and handles great.
