Will these work on a Ruger American rimfire stainless barrel that came with a Picatinny rail?

The Talley 1 in Ruger American Rimfire Low rings do not attach to a picatinny rail. They are designed to fit to the receiver of the Ruger American Rimfire Rifle.

Will a 3x9x40 fit with these mounts?

I would recommend a medium ring with a 40mm objective on that rifle. Item 940758.

Will the low mount work on a 35mm scope or do I need the medium?

I have a Leupold 2x7x33. Will the low mounts clear the front sight notch?

Yes. The low height will offer enough clearance for a 33mm objective scope.
